Assistance Listing Number(s) Funding Opportunity Purpose The purpose of this Notice of Funding Opportunity (NOFO) is to conduct secondary data analyses using existing data collected from humans participating in vision-related clinical trials, epidemiologic, and other clinical research studies or represented in clinical and health care database resources (e.g., electronic health records).
This NOFO may be used to develop new statistical methodologies or test hypotheses using existing data, but this NOFO must not be used to support the collection of new data. This NOFO encourages applications from institutions/organizations that propose to conduct vision-related secondary analyses of data collected from humans. Applications may be related to, but must be distinct from, the specific aims of the original data collection.
The NEI supports an extensive portfolio of clinical trials and large-scale epidemiologic research projects wherein numerous data collection activities are required to meet each project's specific aims.
The resultant wealth of data generated by these studies often provides unique, cost-effective opportunities to investigate additional research questions or develop new analytical approaches secondary to a project's originally intended purpose. Data are not limited to those collected under NEI support, but such data are of the highest programmatic interest.
Funding Opportunity Goal(s) 1) To support eye and vision research projects that address the leading causes of blindness and impaired vision in the U.S. These include retinal diseases; corneal diseases; cataract; glaucoma and optic neuropathies; strabismus; amblyopia; and low vision and blindness rehabilitation.
2) To increase understanding of the normal development and function of the visual system in order to better prevent, diagnose, and treat sight-threatening conditions; and, to enhance the rehabilitation, training, and quality of life of individuals who are partially-sighted or blind.
3) To support a broad program of basic vision research through grants and cooperative agreements; to encourage high quality clinical research, including clinical trials, other epidemiological studies, and health services research; to encourage research training and career development in the sciences related to vision; and to sponsor scientific workshops in high priority research areas to encourage exchange of information among scientists.

Notice of Funding Opportunity Description The goal of this Notice of Funding Opportunity (NOFO) reissuance is to stimulate secondary analyses of data collected from humans to investigate new scientific ideas, models, systems, tools, or methods that have the potential for significant impact in areas relevant to the NEI mission.
Applications may be related to, but must be distinct from, the specific aims of the original data collection. This NOFO will not support the collection of new data, nor analyses of animal data. The NEI supports an extensive portfolio of clinical trials and large-scale epidemiologic research projects wherein numerous data collection activities are required to meet each project's specific aims.
The resultant wealth of data generated by these studies often provides unique, cost-effective opportunities to investigate additional research questions or develop new analytical approaches secondary to a project's originally intended purpose.
A typical project supported under this NOFO is expected to make substantial progress towards having at least one vision-related manuscript submitted to a peer-reviewed journal within the first year.
Examples of secondary analyses projects include, but are not limited to: exploration of associations between risk factors and vision health outcomes development and/or validation of screening algorithms studies related to disease etiology, detection, diagnosis, treatment, progression, exacerbation, endotypes, risk, prevention, and survivorship studies of health disparities identification of novel biomedical and environmental exposures derivation of new biomedical outcomes implementation and translation of research findings into practice development and validation of new statistical methodologies and computational modeling Standardized Ocular Imaging Expectations: The NEI encourages applicants to use common formats for files and metadata standards when incorporating ocular imaging in their research proposals (see NOT-EY-24-006 ).
This applies to both clinical and nonclinical research applications submitted in response to NOFOs where NEI is the issuing or participating organization.
It is anticipated the widespread adoption of standardized imaging formats will significantly advance the field by 1) simplifying streamlined digital workflows; 2) enhancing interoperability of large datasets; and 3) developing extensive and comprehensive training sets for artificial intelligence driven research.
By promoting these standards, the NEI goal is to create an environment where data can be seamlessly integrated and analyzed across different studies, thereby, amplifying the overall impact of vision research. To facilitate the exchange of health care information electronically the NEI encourages integration of FHIR (Fast Healthcare Interoperability Resources) ( https://hl7. org/fhir/us/cdmh/ ), OMOP ( https://www.

Applications Not Responsive to this NOFO The following types of applications will be considered non-responsive to this NOFO and will not be reviewed and will be withdrawn: Studies that propose to collect or generate new data, e.g., grading images, running lab assays Studies that propose to analyze data obtained from non-human samples or animal studies Studies that propose to carry out or complete currently ongoing data analysis or to maintain and distribute data sets.

Funds Available and Anticipated Number of Awards The number of awards is contingent upon NIH appropriations and the submission of a sufficient number of meritorious applications.
The NEI permits direct costs up to $275,000 for the entire two-year project period, plus applicable F&A for consortia. No more than $200,000 in direct costs may be requested in any single year. The total project period for an application submitted in response to this funding opportunity may not exceed two years.

Provide details of the existing data (data sets or databases) and describe how these data will be utilized to address the project's specific aims and NEIs mission. Application Review Information Only the review criteria described below will be considered in the review process. Applications submitted to the NIH in support of the NIH mission are evaluated for scientific and technical merit through the NIH peer review system.
Reviewers will provide an overall impact score to reflect their assessment of the likelihood for the project to exert a sustained, powerful influence on the research field(s) involved, in consideration of the following scored review criteria and additional review criteria (as applicable for the project proposed). An application does not need to be strong in all categories to be judged likely to have a major scientific impact.
Reviewers will consider Factors 1, 2 and 3 in the determination of scientific merit, and in providing an overall impact score. In addition, Factors 1 and 2 will each receive a separate factor score. Factor 1.
Importance of the Research (Significance and Innovation) Evaluate the importance of the proposed research in the context of current scientific challenges and opportunities, either for advancing knowledge within the field, or more broadly. Assess whether the application addresses an important gap in knowledge in the field, would solve a critical problem, or create a valuable conceptual or technical advance.
Evaluate the rationale for undertaking the study, the rigor of the scientific background for the work (e.g., prior literature and/or preliminary data) and whether the scientific background justifies the proposed study. Evaluate the extent to which innovation influences the importance of undertaking the proposed research.
Note that while technical or conceptual innovation can influence the importance of the proposed research, a project that is not applying novel concepts or approaches may be of critical importance for the field. Evaluate whether the proposed work applies novel concepts, methods or technologies or uses existing concepts, methods, technologies in novel ways, to enhance the overall impact of the project. Factor 2.
Rigor and Feasibility (Approach) Evaluate the scientific quality of the proposed work. Evaluate the likelihood that compelling, reproducible findings will result (rigor) and assess whether the proposed studies can be done well and within the timeframes proposed (feasibility). Evaluate the potential to produce unbiased, reproducible, robust data.
Evaluate the rigor of experimental design and whether appropriate controls are in place. Evaluate whether the sample size is sufficient and well-justified. Assess the quality of the plans for analysis, interpretation, and reporting of results.
Evaluate whether the investigators presented adequate plans to address relevant biological variables, such as sex or age, in the design, analysis, and reporting. For applications involving human subjects or vertebrate animals, also evaluate: the rigor of the intervention or study manipulation (if applicable to the study design). whether outcome variables are justified.
whether the results will be generalizable or, in the case of a rare disease/special group, relevant to the particular subgroup. whether the sample is appropriate and sufficiently diverse to address the proposed question(s). For applications involving human subjects, including clinical trials, assess the adequacy of inclusion plans as appropriate for the scientific goals of the research.
Considerations of appropriateness may include disease/condition/behavior incidence, prevalence, or population burden, population representation, and/or current state of the science. Evaluate whether the proposed approach is sound and achievable, including plans to address problems or new challenges that emerge in the work.
For proposed studies in which feasibility may be less certain, evaluate whether the uncertainty is balanced by the potential for major advances. For applications involving human subjects, including clinical trials, evaluate the adequacy and feasibility of the plan to recruit and retain a study population that appropriately models the target population.
Additionally, evaluate the likelihood of successfully achieving the proposed enrollment based on age, race, ethnicity, and sex. For clinical trial applications, evaluate whether the study timeline and milestones are feasible.
Assess whether data are of sufficient quality and in analyzable format to address the specific aims of the study and how well the research plan include plans to control or account for potential biases and deficiencies (e.g., missing, inconsistent or implausible data; misclassification and information bias; selection bias; potential confounding). Factor 3.
Expertise and Resources (Investigator(s) and Environment) Evaluate whether the investigator(s) have demonstrated background, training, and expertise, as appropriate for their career stage, to conduct the proposed work. For Multiple Principal Investigator (MPI) applications, assess the quality of the leadership plan to facilitate coordination and collaboration.
Evaluate whether the institutional resources are appropriate to ensure the successful execution of the proposed work. Additional Review Criteria As applicable for the project proposed, reviewers will consider the following additional items while determining scientific and technical merit, but will not give criterion scores for these items, and should consider them in providing an overall impact score.
Protections for Human Subjects For research that involves human subjects but does not involve one of the categories of research that are exempt under 45 CFR Part 46, evaluate the justification for involvement of human subjects and the proposed protections from research risk relating to their participation according to the following five review criteria: 1) risk to subjects; 2) adequacy of protection against risks; 3) potential benefits to the subjects and others; 4) importance of the knowledge to be gained; and 5) data and safety monitoring for clinical trials.
